Bị NASA cắt nguồn viện trợ, v́ lợi ích nhân loại, tiến sĩ Reed Richards (Ioan Gruffudd) đă cầu cứu sự tài trợ của tỷ phú công nghiệp Victor Von Doom (Julian McMahon).
Reed dẫn đầu một nhóm các nhà khoa học gồm phi hành gia Ben Grimm (Michael Chiklis), Sue Storm (Jessica Alba) - người phụ trách chương tŕnh nghiên cứu về di truyền, vốn là bạn gái cũ của Reed trước đây, phi công Johny Storm (Chris Evans), em trai Sue và Victor - người tài trợ, cũng là một đối thủ trong t́nh yêu của anh - cùng ra ngoài không gian để thực hiện thí nghiệm giải mă bí mật hệ di truyền của con người.
Từ một tính toán sai lầm của Reed về thời gian tiếp xúc với đám mây phóng xạ nên toàn bộ nhóm nghiên cứu sau khi trở về trái đất đă bị biến đổi gen. Họ trở thành những người có khả năng kỳ lạ. Reed có thể kéo giăn bất kỳ bộ phận nào trên cơ thể (Mr. Fantastic). Sue có khả năng tàng h́nh và phát ra năng lượng (Invisible Woman). Johnny có khả năng tự bốc cháy và bay lên cao (Human Torch). Ben bị biến dạng thành người đá cứng rắn và có sức mạnh. Họ đă tập hợp thành bộ tứ giúp đỡ và bảo vệ thành phố New York chống lại Victor. Tên này sau khi nhiễm xạ đă sử dụng sức mạnh của ḿnh phục vụ mục đích cá nhân.
